Fixing A Broken Heart

By Esco Healthcare 29 May, 2018

Stem cells are the most promising and most exciting area in medicine. Stem cells are unspecialized cells capable of renewing themselves through cell division. It can be induced to embody cells with special functions. Due to their potential in regenerating and repairing damaged tissues, scientists are exploring and optimizing stem cells abilities to treat different types of diseases.

One of the treatments with promising result is the optimization of adipose-derived stem cells to treat cardiovascular. According to the World Health Organization (WHO), up to 7.4 million people died of heart attack due to ischemic heart disease.

Ischemic heart disease results from coronary artery disease that eventually lead to myocardial infarction (MI). The cardiomyocytes (cardiac muscle cells) are terminally differentiated cells that has no capability to repair damaged tissues of the heart. With regards to this, scientists find an effective treatment for MI by inducing new adipose-derived stem cells (ADSCs) to differentiate into new cardiomyocytes.

ADSCs can differentiate into cardiomyocytes, endothelial cells and vascular smooth muscle cells. With the use of trans differentiation or cell-diffusion method. They can also contribute towards improvement in cardiac function when these cells are transplanted into a patient with infarcted myocardium. Adipose-derived stem cells have two main biological pathways in treating this type of disease: have anti-inflammatory properties and secretion of repair and growth promoting molecules. Moreover, adipose-derived stem cells exhibit immunomodulatory properties which include the regulation of immune cells.

In this way, these ADCs can differentiate and regenerate into new cardiomyocytes to replace the damage tissues. ADSCs can be extracted via liposuction from subcutaneous fat tissue from a healthy donor, followed by the differentiation method before making a cell bank. Afterwards, these adipose derived stem cells will be expanded in a cell culture.

In this field, many adipose-derived stem cells are needed to meet the required target number of cells. Bioreactors are used for cell expansion.

Esco VacciXcell bioreactors are packed-bed bioreactors that can reproduce these adipose-derived stem cells in large number because of high surface area from the three-dimensional cell culture (BioNOC™ II).

These macroporous carriers are perfect for supporting the growth and proliferation of these adherent cells. these provide suitable environment for ADSC culture that offers better cell-cell communication and let the cells grow at a higher yield. Moreover, these bioreactors operate under Tide Motion principle that enables 100% oxygen and media exchange perfect environment for the adipose-derived stem cells.

Adherent cell culture processing requires more steps and time than suspension. Tide Motion bioreactors offer linearly scalable beginning with research and development up to commercialization to reduce bioprocessing costs and time. Moreover, TideCell® Cell Harvesting System is a powerful and automated system for harvesting viable cells from 2L to 20L TideCell ® bioreactors. Being a closed system, it is perfect for harvesting seed cells for scaling up without having to worry about contamination. The system can harvest up to 1011 cells within 3 hours of operation.

Overall, this technology meets high productivity manufacturing, reproducibility and compliance to GMP standards also it maintains the comparability of the products released and keeping the cost of goods under tight control.

About Myocardial Infarction

Myocardial Infarction (MI) occurs when an area of the heart does not receive enough blood flow and is subsequently damaged or dies. An MI is commonly referred to as a heart attack and is always a medical emergency. Many times, an MI occurs when one of the main arteries to the heart, specifically the coronary arteries, become blocked due a buildup of plaque.

About Tide Motion Bioreactors

Tide Motion pertains to the gentle oscillation of culture medium into and out of the matrix vessel that intermittently exposes the cells to aeration and nutrition. The upward oscillation exposes the cells to nutrition, while the downward oscillation exposes the cells to aeration. At the same time, this process washes away products and wastes. This oscillation produces no air bubbles and low shear stress. View a range of products at http://www.vaccixcell.com/tide-technology/
